چکیده
Dermal fibroblasts and cutaneous nerves are important players in skin diseases, while their reciprocal roles during inflammation have not been characterized. Here we identified an inflammation-induced subset of papillary that promoted aberrant neurite outgrowth psoriasiform by secreting the extracellular matrix protein tenascin-C (TNC). Single-cell analysis fibroblast lineages revealed aTnc+papillary with pro-axonogenesis neuro-regulation transcriptomic hallmarks. TNC overexpression boosted co-cultured neurons, fibroblast-specific ablation suppressed hyperinnervation, immune cell infiltration, alleviated a psoriasis mouse model. γδT cells, main producers type 17 pathogenic cytokines, frequently contacted nerve fibers lesions were likely modulated postsynaptic signals. Overall, our results highlight role inflammation-responsive facilitating neuro-immune synapse formation suggest potential avenues for future therapeutic research.

Cutaneous Leishmaniasis Induces a Transmissible Dysbiotic Skin Microbiota that Promotes Skin Inflammation.
Skin microbiota can impact allergic and autoimmune responses, wound healing, and anti-microbial defense. We investigated the role of skin microbiota in cutaneous leishmaniasis and found that human patients infected with Leishmania braziliensis develop dysbiotic skin microbiota, characterized by increases in the abundance of Staphylococcus and/or Streptococcus. Mice infected with L. major exhibi...
متن کاملHuman tumors instigate granulin-expressing hematopoietic cells that promote malignancy by activating stromal fibroblasts in mice.
Systemic instigation is a process by which endocrine signals sent from certain tumors (instigators) stimulate BM cells (BMCs), which are mobilized into the circulation and subsequently foster the growth of otherwise indolent carcinoma cells (responders) residing at distant anatomical sites. The identity of the BMCs and their specific contribution or contributions to responder tumor growth have ...
متن کاملChronic Skin-Specific Inflammation Promotes Vascular Inflammation and Thrombosis
Patients with psoriasis have systemic and vascular inflammation and are at increased risk for myocardial infarction, stroke, and cardiovascular death. However, the underlying mechanism(s) mediating the link between psoriasis and vascular disease is incompletely defined. This study sought to determine whether chronic skin-specific inflammation has the capacity to promote vascular inflammation an...
متن کاملDownregulation of tenascin expression by glucocorticoids in bone marrow stromal cells and in fibroblasts
Tenascin, a predominantly mesenchymal extracellular matrix (ECM) glycoprotein has a rather restricted tissue distribution, but until now factors that inhibit its expression have not been identified. Glucocorticoids are known to be beneficial for establishment of myelopoiesis in long-term bone marrow cultures. Tenascin was found to be expressed in the bone marrow, and glucocorticoids were found ...
متن کاملThe distribution of tenascin-X is distinct and often reciprocal to that of tenascin-C
We have isolated a cDNA encoding mouse tenascin-X (TN-X), a new member of the family of tenascin genes. The TN-X gene lies in the major histocompatibility complex (MHC) class III region, as it is the case for its human counterpart. On Northern blots we detected a TN-X mRNA of approximately 13 kb in most tissues analyzed, whereas in various mouse cell lines mRNAs of approximately 11 and 13 kb we...
